Field testing of a novel concept for automated on-line monitoring of β-D-glucuronidase activity in water resources

Background and aim of research
Direct determination of ectocellular ß-D-glucuronidase activity (GLUC) has been demonstrated as a potential, cultivation-independent, alternative method for the detection of faecal pollution in different water resources (Fiksdal 2008). Here a novel concept for an automated device capable of measuring the GLUC, after filtration of defined volumes of water, was tested. The main focus of the study was the evaluation of the operational applicability, stability of measurement under different field conditions and reliability of data transfer of the newly developed measuring device. Throughout a time period of two years (2010 to 2011) continuous on-line GLUC data were successfully generated and analysed together with concurrently recorded physicochemical, hydrological and microbiological parameters from two hydro-geological contrasting water resources.
