Production, purification and characterization of invertase by Aspergillus flavus using fruit peel waste as substrate

A. flavus produced high levels of invertase under optimized culture conditions on 4th day of incubation at an optimum pH 5.0, temperature 300C, inoculum size 3% in Czapek Dox using fruit peel waste as a substrate by SmF. Enhanced production occurred on addition of sucrose and yeast extract as nutritional factors. The enzyme was purified to 5.8 fold with recovery of 3.2% by DEAE-column chromatography and the molecular weight was estimated to be 67 KDa by SDS-PAGE. It has a Vmax value of 15.8 U/mg and Km value of 0.23 mg/ml at pH of 6. The enzyme activity was found to be stable at 500C for 30 minutes and it was stimulated by metal ions like Na+ and Ca2+ and inhibited by Zinc.
