This test is intended to detect LMV in suspicious lettuce leaf samples. This virus spreads easily by seeds and aphid vectors. It is most important on lettuce. The symptoms include necrotic or chlorotic local lesions and streaking, then mosaic, vein yellowing, and leaf malformation. For seed testing we recommend using the DAS-ELISA format (BIOREBA Art. No. 160512/160522). The AgriStrip for LMV detection does not work with seeds.

The reagents used for this AgriStrip were made against an isolate of LMV, originally from the Salinas Valley, California, USA (R.L. Gilbertson, personal communication), maintained at Agroscope ACW (Changins, Switzerland) under isolate number LMV 1070. This test specifically reacts with LMV. Strongest coloration of testbands was obtained with a 1:10 to 1:40 (w/v) dilution of an extract of infected lettuce leaves.
Instructions for use
- Place approx. 0.3 g of leaf (≈ 10 cm2) into an extraction bag (Fig. 1*) and add 3 ml of AgriStrip extraction buffer B with a disposable pipette (1:10 w/v).
- Homogenize the tissue with a handheld homogenizer thoroughly for about 4 seconds (Fig. 2*).
- Transfer 4 drops of extract (approx. 150 μl) into a cuvette (Fig. 3*).
- Insert the end of the strip marked «sample» into the extract (Fig. 4*) and observe formation of colored bands.
