Wine Analysis a Unique System From Preparation to Measurement Application Presentation
Wine analysis A unique system from preparation to measurementDidier ConseilThermo Fisher Scientific - FranceJuly 20102The customer and a challenge for the team • The customer • Institut de la vigne et du vin – Gruissan – France: Marie-Agnès Ducasse• Quality control of white and red wines• Her problem: a large number of samples to manage – sampling or addition of a reactive (or dilution) and sampling – wine color determination,… – data export – budget: 25K€ no more… for an automatic system (dilutor, sampler, UV-vis spectrophotometer, PC and software)• The team: a combination of experts • A Thermo Fisher Scientific sales person: Didier Bourgeois• His objective: he must build the best configuration for the best price• A Thermo Fisher Scientific service person: Rabah Rabahi• His challenge: he is not familiar with the system, but he must install it and train our user• A Cetac sales person: Jean Yves Henry (Ambrosia Technologies - France)• He will be present at his step in the process• A software editor: W. Liekmeier (Ascany company - Germany) • His personal support: “the project is on line with our development on the wine analysis market: our native OptLab-SPX software can be improved according to the request of the user”3The CETAC and Ascanis software packages• CETAC software• Control the dilution or the addition of a reactive • Ascanis OptLab-SPX auto software (modified version)• Control the auto sampler/sipper to automatically transfer the samples to the spectrophotometer• Record the sample spectrum • Perform the calculations (color intensity: 420/520, 420/450/620…)• Generate CIE Lab values• Easily export the spectrum into Excel® using the copy/paste command • The exported spectrum are used for MCA (main compound analysis) calculation4Offline dilution sample system• CETAC SDS-550 offline dilution sample system• Auto sampler ASX520 + dilutor + CETAC software• Dilution ratios: • From 1:1 to 1:100 or more with adapted tubes• Rack configurations• Without dilution/additive of samples- Samples: racks 1,2,3 et 4 (360 x 7ml-13mm tubes or 240 x 13ml-16mm tubes)• With dilution/addition of samples- Samples: racks 1 and 2 (180 x 7ml-13mm tubes or 120 x 13ml-16mm tubes) – Diluted samples: rack 3 and 4 (180 x 7ml-13mm tubes or 120 x 13ml-16mm tubes)• P/N: SDS-99-0002BSamplesDiluted Samplesor sample with reactive5Auto sampling and measurement system• Thermo Scientific Evolution 300 UV-Visible spectrophotometer (PC model) with SmartSipper accessory and flow through cell• Evolution™ 300 PC version w/o software (P/N: 10300501)• Smart™Sipper (P/N: 10010401) • 10mm and 1mm flow through cell from Hellma®, Starna®…• 1mm o.p.: red wine• 10mm o.p.: white wine• Note: Thermo Scientific Evolution 160 UV-Vis spectrophotometer can be used with SuperSipper™ accessory• Ascanis OptLab-SPX auto software6Direct measurement on raw sample• Without dilution or addition of a reactive – raw sample• The sipper of the UV-visible spectrophotometer transfers the sample from the auto sampler to the flow through cell• Raw samples are positioned on racks 1, 2, 3 and 4 • Ascanis OptLab-SPX auto software controls the auto sampler, sipper and UV-vis spectrophotometerAutoSampler Sipper AutoSampler Flow through cell – UV vis spectrophotometer7With dilution/addition (1)• With dilution – diluted sample• Dilution: the dilution volume is larger than the sample volume• Ex: dilution for determination of total polyphenols index (IPT) and free antocyans (total pigments content) - The sample is diluted/treated with HCl: 0.1ml wine in 9.9ml HCl (1:100)• 100x dilution allows to read Abs?280nm (IPT = Abs?280nm x dilution factor)• HCl allows to read Abs?520nm - Acid (pH <1) converts anthocyans -> flavylium and gives a red color (free anthocyanes = Abs?520nm x 22.76 x dilution factor mg/l)1/ The syringe pump of the dilutor sips 9.9ml HCl (stock tank) then 0.1ml wine (Racks 1 or 2) 2/ The syringe pump flushes out HCl and wine (racks 3 and 4)• CETAC software controls the dilution8With dilution/addition (2)• With addition of a reactive – sample with a reactive• Addition: the reactive volume is much smaller than the sample volume• Ex: addition of SO2 for determination of resistant pigment to bleaching sulfite- 150µl SO2 is added at 9.85ml sample before reading Abs?520nm1/ the syringe pump of the dilutor sips 150µl SO2 (reactive) then 9.85ml wine (sample) (rack 1 or 2) 2/ The dilutor flushes out sample and reactive (or additive) to rack 3 or 4***• CETAC software controls the addition of reactive• Transfer: • the sipper of the UV-Visible spectrophotometer transfers the sample from the auto sampler to the flow through cell 9With dilution/addition (3)Sipper Raw samples (racks 1 and 2)Treated samples (racks 3 and 4)Flow through cell UV vis spectrophotometerDilutor HCl tankSO2 tank• Remarks: • Sample with its reactive must be manually mixed with a Vortex for a better homogeneity- *** the sample volume (or additive) is flushed out of the pipe before the reactive. A small volume is ejected in a large one. This Cetac configuration cannot be changed.• All samples must at first be diluted/treated before transfer to the trough flow cell• The pipe of the auto sampler must be connected at the dilutor for the dilution/treatment and then at the sipper for the transfer. A manual three way valve can be used. Manual three way valve10The complete system Wine analysis ?A unique system from preparation to measurement The customer and a challenge for the team The CETAC and Ascanis software packages Offline dilution sample system Auto sampling and measurement system Direct measurement on raw sample With dilution/addition (1) With dilution/addition (2) With dilution/addition (3) The complete system
